So this is a curious thing I’ve just learned about. Some of you have heard that COVID19 is mutating more slowly than expected. It turns out that COVID has instructions for creating a proofreading enzyme called ExoN that keeps bad copies (mutations) from being created. This kind of proofreading system is something that happens with our DNA, but is extremely rare in RNA viruses.

Why does COVID do this? Probably because it’s an unusually complicated virus. Its sequence is~30,000 bases long (i.e. 30,000 lines of code). The flu is ~13,000 bases long, HIV is ~9,000. Longer sequence means a bigger chance of a mistake during copying, so COVID has evolved this ExoN proofreading enzyme.

This is important for two reasons: First, if it’s not mutating quickly, that makes it much easier to develop a vaccine that gives lasting immunity. It’s hard to create a vaccine for most RNA viruses because they’re always switching their game up. Also, if we can figure out some kind of drug that suppresses this ExoN proofreading protein, it might cripple the virus’ ability to reproduce itself. The drug remdesivir is supposed to work by targeting this enzyme.

I think we should be cautious. This is apparently something the sciencebros have known about coronaviruses for a long time (the Nature article I linked to was published in 2013!) but it’s new info for me and maybe some of you. Other coronaviruses in the past like SARS and MERS fizzled out on their own before we really had a chance to fully develop tools to fight them. It could be that this COVID-19 thing forces us to create weapons that stop this ExoN enzyme. Or maybe it’s a dead end.

The state of Victoria is getting something like 20 new cases a day, many of which are community transmission. This is sufficient for the state government there to cancel reopening plans and re-impose some restrictions. No more than 20 people in a single space and no more than 5 visitors to households. There’s talk of a harsher lockdown in suburbs where community transmission is taking place.

Nineteen new positive tests were recorded in the past 24 hours. Health Minister Jenny Mikakos announced today that the state’s infectious total has reached 1836 cases, of which 121 remain active.

The 19 new cases follow rises of 25, 13, 18 and 21 cases over the previous four days.

The rise in coronavirus cases has led the government to extend the state of emergency for four more weeks until July 19. A raft of eased restrictions that were due to come into place on Monday have been axed. Police have been instructed to fine those caught breaking the rules.

“It is still a very serious situation,” Ms Mikakos said.

“We have had particularly some concerns around family gatherings, extended family members across many households, visiting each other even when they have been exhibiting mild symptoms and very concerningly even going to work when they had mild symptoms.

“The persistent rise of COVID-19 cases in Victoria means people must once again cancel social plans and struggling businesses must wait.”

My state of South Australia had planned to reopen its borders to all states on July 20 but that may be put on hold as Victoria tries to get its community transmission problems under control. This has owned a friend of mine for a second time. He had a 6-week family holiday planned to the US leaving late March and cancelled in early March as it became clear how bad things were getting. Now he’s booked another family trip to go skiing in the Australian Alps in Victoria, returning July 21 since the government had declared that borders would be open. Looking like a good chance he’ll be cancelling that too.
