If we are discussing music theory, each note has a distinct harmonic. E# doesn’t sound the same as Fb because it literally isn’t the same pitch.
If you mean how we can manifest a note in reality, then yes we are dependent on the note an instrument can physically produce.
Having said that, you will also see this manifest in played music. If you measure the harmonics of a voice, for example, you will note a distinction between the harmonic for E# vs Fb.
